Abstract:
In present study, among different stains used for malaria identification, JSB, Geimsa’s and Leishman’s stains were found to be best for routine diagnosis of malaria, .But there is the problem associated with them is possibility of washing up the parasite from the smear during dehaemoglobination and flushing, which are a common steps in staining procedure.
In contrast, Brilliant cresyl blue dye ( a reticulocyte stain) adapted to stain and identify malaria parasites in whole blood of confirmed malaria had shown better results than other stains more particularly for schizont and ring stages of the parasites. Staining of slides with this stain consumed less time in whole process of malaria diagnosis, as compared to other routine stains used by malaria personels, in primary health centers( PHCs). In addition , there was a clear difference between infected and uninfected RBCs as later remained unstained making identification of infected cell more easier , which is not as such for other stains. Brilliant cresyl blue dye is also useful in scanty parasitemia. The stain had shown higher reliability to ring and schizont stage, while its reliability to gametocyte stage particularly for Plasmodium falciparum was less or not reliable, hence the cytoplasm and the outer membrane of gametocyte remain unstained.
Hence, a new stain ( Brilliant cresyl blue dye ) was adapted to identify malarial parasite, to overcome those limitations associated with conventional stains, therby, rapid diagnosis can be carried out.
